ABSTRACT

Objective To investigate the effect of evodiamine on proliferation of HCT-116 in balb/c nude mice, and to explore possible mechanism. Methods HCT-116 cells were injected into the right armpit of 4 week old Balb/c nude mice, the feeding had been executed at the time of the diameter of the xenografted tumor reached 0.5 cm,at the dose of 3 mg/kg (Evo), body weight and tumor diameter had been tested every three days, and made the curve of body weight and tumor diameter of the mice. All the mice were sacrificed after 22 days of feed-ing,and harvested the xenografted tumors. The morphological difference of the two groups tumor were identified by HE staining, the expression of HDAC3, NF-κB and p53 protein were detected by IHC and Western blot. Results Compared with the control groups, the volume and weight of the tumors in Evo groups were significantly lighter, and the body weight of the nude mice were heavier, the tumor cells in Evo groups were shrink, deeply staining nu-celus and their abnormal mitoses were fewer, the expression of NF-κB and p53 were increased but HDAC3 was de-creased in xenografted tumors treated with Evo(P<0.05). Conclusions Evo can change the expression of NF-κB and p53 by down-regulating HDAC3,and inhibit the proliferation of HCT-116 cells line in vivo.

ABSTRACT

Objective To observe the effect of curcumin on proliferation, apoptosis, Caspase-3 activity and telomerase activity of chronic granulocytic breast cancer SKBR3 cell line, and to investigate the mechanism of curcumin. Methods The cell growth curve was drawn by MTT;the apoptotic cells were detected by Annexin, V and PI double staining; the expression of Caspase-3 was detected by Westernblot; the activity of telomerase was detected by TRAP-PCR silver staining. Results Curcumin could significantly inhibit the proliferation of SKBR3 cells in a time and concentration dependent, has a certain effect on reducing the telomerase activity of SKBR3 cells can enhance the expression of Caspase-3 induced apoptosis. Conclusion Curcumin can effectively inhibit the proliferation of SKBR3 cells, increase the synthesis of Caspase-3 protein, inhibit the activity of telomerase and mediate the apoptosis of SKBR3 cells.

ABSTRACT

Isocorydine and its analogs were extracted from Dicranostigma leptopodum and Stephania yunnanensis through the method of natural products chemistry. Its derivatives were prepared by chemical structure modifications from isocorydine. MTT method was used to study the inhibitory effect of those compounds on the growth of HepG2, HeLa and MGC-803 cancer cell lines in vitro. The results showed that isocorydine and its analogs all have the growth inhibition for those cancer cell lines. This paper investigated the structure-activity relationship of isocorydine and its derivatives with anticancer activity in the aspect of stereochemical structure, functional groups positions of the compounds and the electron density of aromatic rings based on the single crystal diffraction structure and the molecular docking of EGFR and isocorydine.

ABSTRACT

Objective: To observe the effect of recombinant human bone morphogenetic protein 2/poly-lactide-co-glycolic acid (rhBMP-2/PLGA) with core decompression on repair of rabbit femoral head necrosis. Method: Bilateral femoral head necrosis models of rabbit were established by steroid injection. A total of 48 rabbits (96 femoral head necrosis) were randomly divided into 4 groups: Group A, control group with12 rabbits, 24 femoral head necrosis; Group B, treated with rhBMP-2/PLGA implantation after core depression, with 12 rabbits, 24 femoral head necrosis; Group C, treated with rhBMP-2 implantation after core depression, with 12 rabbits, 24 femoral head necrosis; Group D treated with core depression group without implantation, with 12 rabbits, 24 femoral head necrosis. All animals were sacrificed after 12 weeks. The ability of repairing bone defect was evaluated by X-ray radiograph. Bone mineral density analysis of the defect regions were used to evaluate the level of ossification. The morphologic change and bone formation was assessed by HE staining. The angiogenesis was evaluated by VEGF immunohistochemistry. Results: The osteogenetic ability and quality of femoral head necrosis in group B were better than those of other groups after 12 weeks by X-ray radiograph and morphologic investigation. And the angiogenesis in group B was better than other groups. Group C had similar osteogenetic quality of femoral head necrosis and angiogenesis with group D. Conclusion: The treatment of rhBMP-2/PLGA implantation after core depression can promote the repair of rabbit femoral head necrosis. It is a promising and efficient synthetic bone material to treat the femoral head necrosis.

ABSTRACT

Transforming technology for semi-synthesized isocorydione from the natural product ofisocorydine was studied. The factors affecting on the reaction yield were investigated. UV spectrophotometry was used to indicate the semi-synthesized yield of isocorydione. The optimum reaction conditions were determined as following: reacting for 12 h in the solution of sodium dihydrogen phosphate at pH 10, the temperature was 25 degrees C and the ratio of isocorydine to Fremy's radical was 1 : 2. Under the optimum conditions, the yield could reach up to 50.0%. The molecular structure of isocorydione was elucidated by X-ray single-crystal diffraction analysis for the first time.

ABSTRACT

Objective To study the effect of diet containing different proportions of porphyra powder on relieving constipation, volatile organic compounds and inhibiting intestinal E. coli. Method Mice were divided into 3 groups: blank group, 2 test groups. After the mice were allotted for different concentrations of porphyra powder diet for 30 d, the body weight of mice, water-holding capacity, the number and weight of feces within 24 h, the feces pH, E. coli counts and the first black defecating time were detected. The odorous compounds in feces were also measured by headspace solid phase microextraction coupled with GC-MS. Results Compared with the blank group, the water absorbed and number and weight of feces were significantly increased(P
